Unless Japaneses synthesizers are more advanced than English ones, that does not sound synthesized at all. I guess the audio could have been edited afterwards but still probably wouldn't come out like that.
It's synthesized, for sure; one of the particularities of Japanese is that it is more or less made up of consonant-open vowel combinations, enabling it to be more convincingly synthesized than, say, English, with its more varied sound-combinations (diphthongs, foreshortenings, etc.). Still, the intonation is still unnatural, as is the way longer syllables are held, and the like. Doing things like this appears to be all the rage these days on nicovideo, though.
